Protective effect of engeletin on H
2
O
2
induced oxidative stress injury in SH-SY5Y cells

Objective
To investigate the protective effect and molecular mechanism of engeletin on SH-SY5Y cells injured by H

.
Methods
SH-SY5Y cells were pretreated with engeletin (5, 10, 20, 40, 80 and 160 μmol·L
-1
) for 12 h. MTT assay was used to detect the effect of engeletin on cell viability after H
2
O
2
induced injury. The effects of engeletin on the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH PX) after H
2
O
2
induced injury were determined by NBT method and NADPH method; the levels of reactive oxygen species (ROS) in SH-SY5Y cells were determined by flow cytometry. SH-SY5Y cells were pretreated with engeletin (20 μmol·L
-1
) for 12 h and incubated with H
2
O
2
for 6 h, 12 h and 24 h, or pretreated with engeletin (5, 10 and 20 μmol·L
-1
) for 12 h and incubated with H
2
O
2
for 12 h. The expressions of Nrf2 and HO-1 were detected by Western blot.
Results
Engeletin could protect SH-SY5Y cells against H
2
O
2
induced injury. Compared with the model group, engeletin could increase the cell viability, significantly increase the levels of SOD and GSH, and reduce the production of ROS
(P
<0.05). Engeletin promoted the transfer of Nrf2 protein from cytoplasm to nucleus, and reached the peak when SH-SY5Y cells were pretreated with engeletin (20 μmol·L
-1
) for 12 h. In the model group of H
2
O
2
induced injury, engeletin promoted the nuclear transfer of Nrf2 in a concentration dependent manner (
P<
0.05), and promoted the expression of HO-1, a downstream protein of Nrf2 (
P<
0.05).
Conclusion
Engeletin protected SH-SY5Y cells from H
2
O
2
induced injury, and its mechanism was related to antioxidation and promoting the expression of Nrf2 and HO-1.

Effect of dexamethasone combined with curcumin on RAW264.7 cell model

Objective
To explore the anti-inflammatory effect of dexamethasone (DEX) combined with curcumin (CUR) on RAW264.7 cells induced by lipopolysaccharide.
Methods
The Griess colorimetric method was used to investigate the effect of DEX and CUR alone and in combination in inhibiting NO secretion. The Chou-Talalay combined index method was used to judge the drug interactions, and screen the synergistic anti-inflammatory ratio of the two drugs. Rhodamine-123 efflux experiment was used to clarify the reversal effect of CUR on DEX multidrug resistance (MDR) under a synergistic compatibility ratio. DEX and CUR were administered according to the screened synergistic anti-inflammatory ratio, DEX and CUR co-loaded nanoparticles (DEX/CUR-NPs) were prepared by emulsification solvent evaporation method, and their pharmaceutical properties and anti-inflammatory activity in vitro were investigated.
Results
When the compatibility molar ratio of DEX and CUR was 1∶5, the CI of each dose was less than 1, showing a synergistic anti-inflammatory effect, and CUR can inhibit the MDR phenomenon of DEX at this ratio. DEX/CUR-NPs was prepared according to the molar ratio of DEX and CUR with 1∶5. The particle size of the DEX/CUR-NPs was (145.5±2.1) nm, the zeta potential was (-20.54±0.98) mV, and the entrapment efficiency of DEX and CUR were (78.9±5.6)% and (96.4±2.4)%, respectively. DEX/CUR-NPs had good sustained-release properties and synergistic anti-inflammatory activity in vitro, and reverse the MDR of DEX.
Conclusion
DEX and CUR can play a good synergistic anti-inflammatory effect under the compatibility ratio of 1∶5. DEX combined with CUR is a potential candidate combination of anti-inflammatory drugs.

Curcumin promotes bone differentiation of rat bone marrow mesenchymal stem cells in inflammatory microenvironment

Objective
To study the effect of curcumin on the osteogenic differentiation of rat bone marrow mesenchymal stem cells (BMSCs) under inflammatory microenvironment.
Methods
BMSCs were isolated and cultured, 10ng/ml TNF-α was used to induce cell inflammation, cells were treated with different concentrations of curcumin, and cell proliferation was detected by CCK-8 to select the appropriate concentration of curcumin for the experiment; the cells were divided into normal group, inflammation group and curcumin+inflammatory group; osteogenic differentiation was dectected with alkaline phosphatase (ALP) and alizarin red staining; the expression of bone sialoprotein (BSP) and runt-related transcription factor 2 (Runx2) was detected by real-time fluorescent quantitative PCR.
Result
Bone marrow mesenchymal stem cells were successfully isolated and cultured; the low concentration of curcumin had no cytotoxicity, and 1μmol /L was the optimum concentration. Inflammation decreased the expression of BSP and runx2, ALP staining and mineralized nodules were reduced; however, curcumin can reverse this trend and increase the expression of BSP and runx2 in the inflammatory state (
P
<0.05), and the ALP staining was deeper and the mineralized nodules increased compared with the inflammatory group.
Conclusion
Curcumin can promote osteogenic differentiation of bone marrow mesenchymal stem cells under inflammatory microenvironment.

Objective

Methods
MIN6 cells were treated with 50ng/ml VEGF-B protein under the conditions of 5.5mM and 25mM glucose. ELISA kit was used to detect insulin secretion, intracellular Ca

, cAMP and ATP content in MIN6 cells at 2 hours and 24 hours. After knocking down VEGF-B in MIN6 cells with siRNA for 48 hours, the insulin secretion, intracellular Ca
2+
, cAMP and ATP content of MIN6 cells were measured at 2 hours and 24 hours under the conditions of 5.5mM and 25mM glucose.
Results
After the intervention of VEGF-B in MIN6 cells, insulin secretion reduced, and the content ofCa
2+
, cAMPand ATP in the cells all decreased. After knocking down VEGF-B with siRNA, the secretion of insulin increased, and the content of Ca
2+
, cAMP and ATP in the cells were consistent with the change trend of insulin.
Conclusion
VEGF-B may affect the insulin secretion of pancreatic β-cells by affecting the content of Ca
2+
, cAMP and ATP.

Identification of differently expressed hub genes in pancreatic cancer by weighted gene co-expression network analysis

Objective
To identify the potential key genes and molecular mechanism of pancreatic cancer by weighted gene co-expression network analysis (WGCNA).
Methods
The gene data of pancreatic cancer and control group were obtained from the cancer genome atlas (TCGA) database. Differentially expressed genes (DEGs) were identified by limma package in R. Then, WGCNA was used to construct the gene co-expression network of pancreatic cancer, identify the co-expression modules, analyze the protein- protein interaction (PPI) and screen the key genes.
Results
Totally 106 DEGs were identified, and a key module (MEpurple) was identified by WGCNA analysis. We further screened 10 key genes, including PKP3, EPCAM, RAB25, CBLC, AP1M2, PRP15L, B3GNT3, ESRP1, AGR2, ARHGEF16.
Conclusion
By using WGCNA algorithm, we have identified the modules and genes related to pancreatic cancer, which provides a theoretical basis for further research on the molecular mechanism of pancreatic cancer development and treatment.

Changes of intestinal microflora in patients with type II diabetic diarrhea based on 16S rDNA high-throughput sequencing

Objective
To study the changes of intestinal microflora in patients with type 2 diabetic diarrhea by 16S rDNA high-throughput sequencing technology.
Methods
Fecal samples of 18 patients with type 2 diabetic diarrhea and 23 healthy participantswere collected to test the intestinal flora genome. The second generation high-throughput gene sequencing technology was used to amplify and sequence the v3-v4 hypervariable region of 16S rDNA, and the bioinformatics analysis was carried out to compare the differences of intestinal flora structure and species between the two groups.
Results
There was no difference in age, gender and body mass index between the two groups. Alpha diversity analysis showed that the abundance and diversity of intestinal flora in patients with type 2 diabetic diarrhea were significantly lower than those in healthy controls,
P
<0.05. At the phylum level, the proportion of Bacteroides in the diarrhea patients with type 2 diabetes was significantly lower than that in the healthy group,
P<
0.05, while Firmicutes and verruca were significantly higher than those in the healthy group, with average
P
<0.05. In addition, the ratio of Firmicutes/Bacteroides in the patient group was significantly higher than those in the healthy group,
P
<0.05. At the genus level, fecobacteria in diarrhea patients with type 2 diabetes was significantly lower than that in healthy control group,
P
<0.05.
Conclusion
16S rDNA high-throughput sequencing technology is helpful to analyze the diversity of intestinal flora in patients with type 2 diabetic diarrhea. This technology may provide novel ideas and theoretical basis for the study of the intestinal flora in patients with type 2 diabetic diarrhea.

Expression and prognostic value of aldehyde dehydrogenase1 A1 and transforming growth factor-β1 in the primary carcinoma and lymph node metastatic carcinoma of breast cancer

Objective
To investigate the expression and prognostic value of aldehyde dehydrogenase 1 A1 (ALDH1A1) and transforming growth factor β1(TGF-β1) in the primary carcinoma and matched lymph node metastatic carcinoma of breast cancer, and to explore the correlation between ALDH1A1 and TGF-β1expression.
Methods
Totally 197 cases of breast invasive carcinoma of no specific type with lymph node metastasis were selected. Immunohistochemistry was used to detect the expression of ALDH1A1 and TGF-β1. The clinical pathological parameters and prognosis were statistically analyzed.
Results
The concordance rate of the ALDH1A1 and TGF-β1expression between primary cancers and paired lymph node metastases were 87.82% and 86.80%, respectively, and there was a significant positive correlation between them (
P
<0.05, respectively). Kaplan-Meier survival analysis combined with Log-rank test showed that the ALDH1A1 and TGF-β1were poor prognostic factors in primary cancer and lymph node metastasis (
P
<0.05). Univariate survival analysis by Cox proportional hazards regression model revealed that the overall survival rates was correlated with histological grade, tumor size, number of positive lymph nodes, ALDH1A1 and TGF-β1 in primary cancer and lymph node metastasis (
P
<0.05). Multivariate analysis showed that ALDH1A1 in primary cancer, TGF-β1 in lymph node metastasis and number of positive lymph nodes were independent prognostic factors (
P
<0.05). Spearman's correlation showed that the expression of ALDH1A1 was significantly related to TGF-β1 in both primary cancer and lymph node metastasis (
P
<0.05).
Conclusion
The protein expression of ALDH1A1 is positively related with TGF-β1 in both primary cancer and lymph node metastasis, and both of them are positively related with poor prognosis. ALDH1A1 in the primary carcinoma and TGF-β1 in lymph node metastatic carcinoma are independent factors for assessing the prognosis of breast carcinoma.

Study on risk factors and rules of lymph node metastasis in non-small cell lung cancer with a diameter of ≤3 cm

Objective
To explore the risk factors and rules of lymph node metastasis in non-small cell lung cancer with a diameter of ≤3 cm, and to provide a theoretical basis for surgical lymph node removal.
Methods
Data of 526 cases of non-small cell lung cancer with a diameter of≤3 cm were collected in the Department of Thoracic Surgery, Binzhou Medical University Hospital from January 2010 to July 2020, andchi-squaretest and logistic regression were used to analyze the risk factors of lymph node metastasis.
Results
Single factor and multiple factor analysis show the pathological type of tumor, tumor diameter, serum CEA, vascular tumor thrombus, and visceral pleura invasion are risk factors for lymph node metastasis, with
P
<0.05.
Conclusion
For non-small cell lung cancer ≤3 cm in diameter,the pathological type of tumor, tumor diameter, serum CEA, vascular tumor thrombus, visceral pleura invasion are importantly related to lymph node metastasis, and when the above risk factors are present, surgery requires systematic lymph node dissection.

Meta-analysis of the efficacy and safety of helmet and face mask noninvasive ventilation in patients with chronic obstructive pulmonary disease

Objective
To compare the efficacy and safety of helmet and face mask noninvasive ventilation in clinical treatment of patients with chronic obstructive pulmonary disease (COPD).
Methods
Retrieval related studies were carried out of COPD patients treated with helmet and face mask noninvasive ventilation in PubMed, Web of Science, CNKI, Wanfang and Weip database and Review Manager 5.3 statistical software was used for meta analysis.
Results
Nine articles were selected with 606 patients including 304 cases in the helmet group and 302 cases in the face mask group. Meta analysis showed that there was no significant difference between helmet and face mask noninvasive ventilation in reducing respiratory frequency, increasing oxygen and index, and reducing partial pressure of carbon dioxide. For patients of 70 years old and below, helmet could decrease respiratory frequency (
P<
0.05) and increase oxygenation index (
P
<0.05). Helmet noninvasive ventilation could reduce the incidence of complications such as facial skin ulcers or necrosis, conjunctivitis, intolerance and air leakage, and the difference was statistically significant (
P
<0.05).
Conclusion
Compared with face mask noninvasive ventilation, helmet noninvasive ventilation can significantly reduce clinical complications in COPD patients. Especially in patients of 70 years old and below, it can improve the oxygenation index and reduce respiratory frequency, having higher efficacy and safety.

Analysis on the changes and drug resistance of multi-drug-resistant bacteria in a hospital

Objective
To understand the species, drug resistance and changes of multi-drug resistant bacteria (MDRO) in a hospital in recent 3 years, so as to provide basis for the prevention and control of nosocomial infection and clinical rational drug use.
Methods
By using the method of retrospective analysis, the detection and drug sensitivity test data of multi-drug resistant bacteria in the samples of inpatients in the hospital from 2017 to 2019 were collected, and the species, drug resistance and changes of multi-drug resistant bacteria were analyzed.
Results
Totally 933 strains of multi-drug-resistant bacteria were detected in our hospital from January 2017 to December 2019. The main sources of multi-drug-resistant bacteria infection samples were sputum (30.12%), followed by urine (30.01%), secretion (28.51%), blood (10.40%) and so on. Gram-negative bacteria accounted for 83.49%, and Gram-positive bacteria accounted for 16.51%. The top 5 strains of 923 strains were Escherichia coli producing extended-spectrum β-lactamase (ESBLs), Klebsiella pneumoniae producing ESBLs, methicillin-resistant Staphylococcus aureus (MRSA), Carbapenem-resistant Acinetobacter baumannii (CR-AB), multidrug-resistant Pseudomonas aeruginosa (MDR-PA). The resistance rate of ESBLs positive multidrug resistant bacteria to penicillins, ceftriaxone and cefazolin was more than 90% ,and the sensitivity to carbapenem, aminoglycosides,cefoperazone/sulbactam was more than 90%, especially to ertapenem (99.4%-100%). The resistance rate of Pseudomonas aeruginosa to ticacillin/clavulanic acid, imipenem and nitrofurantoin was more than 70%, but the sensitivity to tobramycin was 92.9%. Acinetobacter baumannii was highly resistant to cephalosporins, imipenem, aminoglycosides, quinolones and other common antibiotics, with a resistance rate of more than 95% and a sensitivity of 90.2% only to cefoperazone/sulbactam. The resistance rate of MRSA to penicillins was 100%.
Conclusion
The infection of multi-drug resistant bacteria in the hospital is becoming more and more serious in the past three years. Escherichia coli (producing ESBLs) in Gram-negative bacteria decreased year by year, Klebsiella pneumoniae (producing ESBLS) increased at first and then decreased, and Gram-positive bacteria MRSA increased year by year. However, the infection of multi-drug-resistant bacteria in the hospital is still mainly Gram-negative bacteria. We should strengthen the monitoring of multi-drug-resistant bacteria, select antibiotics reasonably.

Effect of reduning injection on mycoplasma pneumonia in children and on IFN-γ, TNF - α and IL-6

Objective
To investigate the clinical efficacy of reduning Injection in the treatment of mycoplasma pneumonia (MPP) in children and the level of IFN-γ, TNF-α and IL-6 in serum.
Methods
One hundred and twenty children with Mycoplasma pneumoniae pneumonia hospitalized in the second ward of Pediatrics, Yidu Central Hospital of Weifang from January 2018 to December 2019 were selected as the research objects. They were randomly divided into the control group (odd numbers as the control group) with 56 cases and the reduning group with 64 cases (even numbers as the reduning group). The control group was given conventional treatment; the reduning group was additionally given reduning 0.5ml/kg.d intravenous drip besides conventional treatment, with the maximum amount of 20 ml daily for 1 week. The clinical manifestations, therapeutic effects and serum inflammatory factors such as IL-6, TNF - α, IFN-γ before and after treatment were compared between the two groups.
Results
The two groups were cured after active treatment, but the length of hospital stay was different. The results showed that the fever subsided time, cough disappearance time, rales disappearance time and hospitalization days in reduning group were lower than those in the control group (
P
<0.05); the effective rate of reduning group was significantly better than that of control group; after 1 week of treatment, the level of IL-6 in reduning group was significantly lower than that in the control group (
P
<0.05). After 2 weeks of treatment, the levels of IL-6, TNF - α and IFN-γ in reduning group were significantly lower than those in the control group (
P
<0.05). There were few adverse reactions in the two groups.
Conclusion
Reduning can effectively improve the clinical manifestations of MPP, inhibit the expression of IL-6, TNF - α, IFN-γ and other inflammatory factors, so as to improve the effective rate of MPP treatment. Reduning is safe, efficient and has less adverse reactions, which is worthy of clinical promotion.

Comparison of value of perilesional and intralesional radiomics features for diagnosis of clinically significant prostate cancer

Objective
To compare value of perilesional volume (PLV) and intralesional volume (ILV) radiomics features for diagnosis of clinically significant prostate cancer.
Methods
One hundred and forty patients who received prostate magnetic resonance imaging (MRI) examination (training set 112, testing set 28) was respectively analyzed. ILV and PLV were segmented on T2 weighted imaging (T2WI), apparent diffusion coefficient (ADC) map, dynamic contrast enhanced magnetic resonance imaging (DCE-MRI), respectively. Radiomic features extracted from ILV and PLV were selected via univariate analysis and least absolute shrinkage and selection operator (LASSO) combined with 10-fold cross validation. ILV and PLV prediction models were built based on LASSO regression. Area under receiver operating characteristic curve (AUC) and DeLong test were used to evaluate and compare the models.
Results
AUC of ILV model in training and testing set were 0.91, 0.91, respectively. AUC of PLV model in training and testing set were 0.89, 0.87, respectively. Prediction ability of two models were not significantly different.
Conclusion
The value of PLV radiomics features was lower than ILV radiomics features for diagnosis of csPCa. There was no significant difference between them.

Methods
The compounds were isolated and purified by TLC, Sephadex LH-20 column, sillica gel column chromatography and semi-preparative HPLC. Their chemical structures were identified by NMR, MS and comparison with the data of literatures.
Results
From the CH3OH extract of
Aaptos suberitoides,
seven compounds were isolated and their structures were determined as 3-(methylamino)demethyl(oxy)aaptamnie, aaptamine, uracil, thymine, cyclo(Pro-Ser), cyclo(Pro-Thr) and thymine nuleoside.
Conclusion
Aaptamine, cyclo(Pro-Ser) and cyclo(Pro-Thr) were isolated from the genus Aaptos for the first time.

Objective
To understand the changes in medical costs brought by the county reform to patients, evaluate the impact of the medical community policy, and clarify the welfare and impact of the medical community reform on the grass-roots people.
Methods
The amount of hospitalization, total medical expenses, reimbursable expenses, actual medical expenses and personal expenses before and after the reform of Funan county medical community were statistically described. The paired

After the comparison of the data before and after the reform, it was found that the amount of hospitalization increased by 26.7% in the year after the reform, the growth rate of the per capita total medical expenses was 9.2% before the reform and 1.8% after the reform, and the growth rate of the per capita total medical expenses was greatly reduced. The real cost per capita of health insurance continued to rise; The annual growth rate of per capita personal expenditure decreased from 12.0% before the reform to 6.7% after the reform. Conclusion
The medical community reform has effectively controlled the rise of personal total medical expenses, increased the input of medical insurance, and brought benefits to grass-roots citizens. The implementation of the medical community reform is conducive to the rational allocation and use of medical resources, reducing the burden of medical care in various localities, and reducing the treatment costs of the people at the grass-roots level, and achieving the purpose of rational use of medical insurance costs.

Analysis of status and influencing factors of cognitive function in adult inpatients with hypertension

Objective
To investigate the status and influencing factors of cognitive function in adult inpatients with hypertension.
Methods
One hundred and fourteen adult inpatients with hypertension in the cardiology department of a tertiary hospital in Shandong province were selected and assessed with the general information questionnaire, Beijing version of the montreal cognitive assessment scale (MoCA).
Results
The total scores of MoCA in adult inpatients with hypertension was (24.37±2.69), and the incidence of cognitive dysfunction was 56.14% (64/114). Correlation analysis showed that the total score of MoCA in adult inpatients with hypertension was significantly negatively correlated with age (

=-0.45) and urea (
r=-0.21) (P
<0.05). Multiple linear regression analysis showed that education level

=0.23) was a protective factor for cognitive function (

Conclusion
The incidence of cognitive dysfunction in adult inpatients with hypertension was high. Educational level, age, hypertension grade, and stroke history were the influencing factors of cognitive function. It is suggested to develop and implement personalized anti-hypertensive treatment programs to effectively control hypertension, prevent or delay the occurrence and development of cognitive dysfunction in adult inpatients with hypertension. At the same time, attention should be paid to the impact of education on the cognitive function of adult inpatients with hypertension.

Effect of acceptance and commitment therapy on negative emotion and self-efficacy in patients with cerebral infarction and hemiplegia

Objective
To investigate the effect of acceptance and commitment therapy on negative emotion and self-efficacy in patients with cerebral infarction and hemiplegia.
Methods
Convenience sampling method was used to select 81 cases of patients with cerebral infarction and hemiplegia in the department of neurology of a tertiary hospital as the research objects. The patients were randomly divided into the control group and the experimental group according to the admission sequence and admission number. There were 41 cases in the control group and 40 cases in the experimental group. The control group received routine care, and the experimental group received acceptance and commitment therapy besides routine care. The negative mood improvement and self-efficacy of the two groups were compared at before and after the intervention.
Results
Two weeks and 4 weeks after ACT intervention, the GAD-7 score of patients with CI hemiplegia in the experimental group was (10.62±1.38) and (8.91±1.81), lower than those in the control group (11.04±1.32) and (10.14±1.84), (average

In recent years, with the development of science and technology, mass spectrometry (MS) has emerged as an innovative analytical technique. MS is suitable for a large number of analyses and is widely used in the field of protein analysis. It has become the main choice for proteomics applications with its efficiency, sensitivity, accuracy, stability and convenience. Proteomics is essential for early stage diagnosis, prognosis and monitoring of disease development. Among them, the protein post-translational modification(PTM) is an important way of regulating protein functionand. The testing of PTM is very important in the experimental process. This article reviews mass spectrometry analysis of PTM(ubiquitination, sumoylation, acetylation, glycosylation)andthe new sumoylation mass spectrometry detection method defined in our laboratory.This article also discusses the application of mass spectrometry in precision medicine.
