Effects of VEGF-B on insulin secretion and Ca2+, cAMP, ATP content of pancreatic β cells |

Abstract Objective To investigate the effects of VEGF-B on insulin secretion and intracellular Ca2+, cAMP and ATP content in pancreatic islet MIN6 cells and explore their possible mechanisms.Methods MIN6 cells were treated with 50ng/ml VEGF-B protein under the conditions of 5.5mM and 25mM glucose. ELISA kit was used to detect insulin secretion, intracellular Ca2+, cAMP and ATP content in MIN6 cells at 2 hours and 24 hours. After knocking down VEGF-B in MIN6 cells with siRNA for 48 hours, the insulin secretion, intracellular Ca2+, cAMP and ATP content of MIN6 cells were measured at 2 hours and 24 hours under the conditions of 5.5mM and 25mM glucose. Results After the intervention of VEGF-B in MIN6 cells, insulin secretion reduced, and the content ofCa2+, cAMPand ATP in the cells all decreased. After knocking down VEGF-B with siRNA, the secretion of insulin increased, and the content of Ca2+, cAMP and ATP in the cells were consistent with the change trend of insulin.Conclusion VEGF-B may affect the insulin secretion of pancreatic β-cells by affecting the content of Ca2+, cAMP and ATP.
