Effects of bisphenol A poisoning on testicular structure and expression of MnSOD-SIRT3 protein in male pups |

Abstract Objective To investigate the effects of bisphenol A (BPA) poisoning on testicular structure and MnSOD-SIRT3 protein expression in male mice.Method ICR pregnant rats were randomly divided into four groups,and since the first day of conception,drinking water was tainted in the solvent control group,the BPA low dose group (10 nmol/L),the BPA dose group (50 nmol/L),and the BPA high dose group (300 nmol/L).Six weeks after birth,the male mice were put to death and the testicular tissues were removed.HE dyed and electron microscope were used to observe the changes in testicular structure,Hoechst 33258 staining was used to observe apoptosis,and Western blot was used to detect the expressions of MnSOD and SIRT3 proteins in testicular tissues.Results Compared with that in the solvent control group,HE staining showed that the morphology of testicular tissues in the dose groups was damaged,which was more serious in the medium and high dose groups.Electron microscope showed that mesenchymal cells,spermatogonial cells and supporting cells were all damaged in the middle and high dose groups.Hoechst 33258 staining showed that apoptosis of cells in testicular tissues of the dose groups was significantly higher than that of the control group.Western blot showed that with the increase of the dose,the expressions of MnSOD and SIRT3 proteins in testicular tissues decreased.Conclusion BPA can affect the histological structure of testis by inducing the apoptosis of spermatogenic cells and mesenchymal cells.The mechanism may be related to the reduction of MnSOD and SIRT3 expressions in testis.
