Relief of the the neuropathic pain with procaine by the inhibition on JAK2/STAT3 signaling

Objective
To investigate the role and the possible mechanism of procaine in relieving neuropathic pain (NPP) of bilateral chronic constriction (CCI) rat.
Methods
Eighteen SD rats were divided into three groups randomly,namely,the sham surgery group,the CCI group and the CCI+procaine group.Rats were pretreated with procaine by intrathecal injection,then NPP rat model was induced by sciatic nerve chronic compression injury (CCI) and behavior tests were performed to analyze the pain behaviors upon mechanical,thermal and cold stimulations.Spinal expression of Janus kinase 2 (JAK2) and activator of transcription 3 (STAT3) was detected by qRT-PCR and western blot.
Results
Procaine pretreatment improved the pain behaviors of model rats upon mechanical,thermal and cold stimulations,with the best effect occurring on the 15th day post model construction.Procaine also inhibited JAK2 and STAT3 expression in both mRNA and protein levels.
Conclusion
These findings indicate that procaine is capable of relieving NPP.Its role may be associated with the inhibition on JAK2/STAT3 signaling.

Change of ALDH2 expression in mouse testicular mesenchymal cells induced by single heat stress

Objective
To investigate the role of ALDH2 in the reversibility of mouse testicular reproductive function impaired by single heat stress.
Methods
Eighteen C57 male mice were randomly divided into the heat stress group and the control group and immersed in water at 43℃ and 25℃,respectively,for 15 minutes.The heat treatment group was divided into 3 subgroups (
n=
3) on day 1,7 and 14 after treatment,and a control group (
n
=3) was set in each group.At 1,7,and 14 days of heat exposure,all the mice were sacrificed and their testis tissues collected for measuring the expression level of the ALDH2 protein by immunohistochemistry and Western blot.
Results
Immunohistochemistry showed that ALDH2 protein was expressed in Leydig's cells.Western blot showed that there was no significant difference in ALDH2 protein expression on the first day after thermal stress compared with the control group.Compared with the control group and the first day of thermal stress,ALDH2 protein expression was significantly increased on day 7 and 14 after thermal stress (

<0.01).There was no significant difference in ALDH2 protein expression between day 7 and Day 14 of thermal stress.
Conclusion
The expression of ALDH2 protein increased significantly in 7 days and 14 days after exposure.

Proliferation and apoptosis in BV-2 cells transfected by pre-hsa-miR-98

Objective
To transfect rat microglia BV-2 cells by pre-hsa-miR-98 plasmids HmiR and CmiR and investigate the transfection efficiency,proliferation inhibition and apoptotic rates.
Methods
The pre-hsa-miR-98 plasmids HmiR and CmiR,from a control vector,were transfected into BV-2 cells.Transfection efficiency was investigated by fluorescence microscopy and flow cytometry.The proliferation in the cells was tested by MTT assay.Apoptotic cells induced by the transfection were measured by TUNEL and flow cytometry.
Results
The rates of transfection in the cells were about 60%,investigated by fluorescence microscopy and flow cytometry respectively.The proliferation rates in the transfected cells measured by MTT assay decreased with increase of the transfection hours.The apoptotic cells of rates among the tested groups were not different significantly.
Conclusion
The pre-hsa-miR-98 plasmids HmiR and CmiR transfected BV-2 cells successfully.The proliferation inhibition in the cells was similar between the two groups.Apoptosis was not significantly induced by the transfection.The recombinant plasmids constructed are useful for the coming researches.

Effects of bisphenol A poisoning on testicular structure and expression of MnSOD-SIRT3 protein in male pups

Objective
To investigate the effects of bisphenol A (BPA) poisoning on testicular structure and MnSOD-SIRT3 protein expression in male mice.
Method
ICR pregnant rats were randomly divided into four groups,and since the first day of conception,drinking water was tainted in the solvent control group,the BPA low dose group (10 nmol/L),the BPA dose group (50 nmol/L),and the BPA high dose group (300 nmol/L).Six weeks after birth,the male mice were put to death and the testicular tissues were removed.HE dyed and electron microscope were used to observe the changes in testicular structure,Hoechst 33258 staining was used to observe apoptosis,and Western blot was used to detect the expressions of MnSOD and SIRT3 proteins in testicular tissues.
Results
Compared with that in the solvent control group,HE staining showed that the morphology of testicular tissues in the dose groups was damaged,which was more serious in the medium and high dose groups.Electron microscope showed that mesenchymal cells,spermatogonial cells and supporting cells were all damaged in the middle and high dose groups.Hoechst 33258 staining showed that apoptosis of cells in testicular tissues of the dose groups was significantly higher than that of the control group.Western blot showed that with the increase of the dose,the expressions of MnSOD and SIRT3 proteins in testicular tissues decreased.
Conclusion
BPA can affect the histological structure of testis by inducing the apoptosis of spermatogenic cells and mesenchymal cells.The mechanism may be related to the reduction of MnSOD and SIRT3 expressions in testis.

Effects of total flavonoids of Gynura formosana Kitam on the proliferation and apoptosis of the human multiple myeloma cell line U266

Objective
To investigate the effects of total flavonoids of Gynura formosana Kitam (TFG) on the growth,proliferation and apoptosis of the human multiple myeloma cell line U266.
Methods
The proliferations of the cells were detected by MTT.Observation was conducted on morphological change and apoptosis of U266 cells treated with for TFG 24h by inverted microscope and fluorescence microscope.The cell cycle and cell apoptosis were detected by flow cytometry.
Results
The MTT assay detected that TFG had dose and time dependent effects on the proliferation inhibition of U266,

<0.05.The apoptosis rates of U266 cells treated with TFG of 100ug/ml increased significantly.The U266 cell cycle stayed in S phase.
Conclusion
TFG could inhibit the proliferation of U266,which may be related to cell cycle capture and induction of apoptosis.

Research on rating of prognostic factors in children immune thrombocytopenia

Objective
To investigate the factors affecting the prognosis of children immune thrombocytopenia,and to establish a clinical scoring system for the prognosis of children immune thrombocytopenia.
Methods
(1)Data of 206 children with immune thrombocytopenic treated in our hospital from March 2016 to February 2019 were statistically analyzed.Gender,age,i-nitial platelet count,bleeding,infection,and therapeutic effect were recorded,respectively.Moreover,the number of megakaryocytes of 103 children who underwent bone marrow biopsy were recorded.(2)Outpatient and telephone follow-up were conducted for the children.(3)To make univariate and multivariate Logistic regression analysis on the prognostic factors.(4)To assign reasonable scores to risk factors and to calculate the relationship between score and prognosis.
Results
Among 206 cases with immune thrombocytopenic,170 cases had a good prognosis and 36 had a poor prognosis;among the 103 cases who underwent bone marrow biopsy,76 cases had good prognosis and 27 cases had poor prognosis.The results of statistical analysis showed that there were statistically significant differences in age,prodromal infection,bleeding,initial platelet count and initial Megakaryocyte count,without statistically significant differences in gender or treatment.In the multivariate Logistic regression analysis,the independent risk factors affecting prognosis were age,bleeding,prodromal infection,initial platelet count and initial Megakaryocyte count.The above factors were assigned and the total score was calculated.The results showed that the lower the score was,the better the prognosis was.
Conclusion
Age,bleeding,prodromal infection,initial platelet count and initial Megakaryocyte count have important predictive significance for the prognosis of immune thrombocytopenia and can be used to guide the treatment.

Effect of lumbar interbody fusion combined with unilateral pedicle screw fixationfor treatment of degenerative lumbar spondylolisthesis

Objective
To evaluate the effect of posterior lumbar interbody fusion combined with unilateral and bilateral pedicle screw fixation in treatment of single-level lumbar degenerative spondylolisthesis.
Methods
Fifty-two patients with single-level lumbar degenerative spondylolisthesis were treated with posterior lumbar interbody fusion combined with unilateral or bilateral pedicle screw fixation.According to the different fixation,they were divided into the unilateral group (25 cases) and the bilateral group (27 cases).The preoperative and postoperative visual analogue scale (VAS) and Oswestry disability index (ODI) scores,operation time,intraoperative blood loss,hospitalization costs and fusion rate were recorded and compared between the two groups.
Results
All patients completed the operation successfully and were followed up.At the last follow-up,VAS and ODI scores were significantly lower than those before surgery,

<0.01.The operation time,intraoperative blood loss and operation cost of patients in the bilateral group significantly increased,compared with those in the unilateral group,

<0.01.At the last follow-up,solid fusion was achieved in both groups.
Conclusion
The posterior lumbar interbody fusion combined with unilateral pedicle screw and bilateral pedicle screw fixation in treatment of single-level lumbar degenerative spondylolisthesis have comparable clinical outcomes,and unilateral pedicle screw has more advantages on operation time,intraoperative blood loss and hospitalization costs.

Repair of dorsalis pedis tendon and skin defect with anterolateral femoral perforator flap combined with fascia lata

Objective
To explore the clinical effect of anterolateral femoral perforator flap combined with fascia lata in repairing the tendon and skin defect of dorsum pedis muscle.
Methods
The clinical data of 12 patients with dorsalis pedis tendon and skin defect in our hospital from January 2014 to March 2018 were analyzed retrospectively.The tendon defect was repaired with fascia lata in 12 patients,and the anterolateral femoral perforator flap was used to cover the wound.
Results
All the 12 flaps survived.The patients were followed up for 6 to 24 months with an average of 10 months.The shape of flaps and shoes'wearing were satisfactory.The sensory function of the flap recovered well,and the distance perception was 10-15 mm at two points.
Conclusion
The anterolateral femoral perforator flap combined with fascia lata is a reliable method for repairing the tendon and skin defect of the dorsum pedis with short course,less trauma to the donor site,satisfactory appearance and function.

Analysis of risk factors of pulmonary infection in the early stage of severe head injury

Objective
To analyze the risk factors of early pulmonary infection in patients with severe craniocerebral injury.
Methods
The clinical data of 300 patients with severe head injury were retrospectively analyzed,and the patients were divided into the non-infection group (147 cases) and the infection group (153 cases) according to the occurrence of pulmonary infection.Univariate analysis and multivariate Logistic regression analysis were used to compare the differences between the two groups.
Results
The incidence of pulmonary infection in 300 patients with severe head injury was 51% (153/300).Multiple factor logistic regression analysis showed that smoking,GCS score and glucocorticoid were independent risk factors of pulmonary infection in patients with severe head injury,and the

) were 0.327 (0.151-0.709),0.292 (0.212-0.403) and 2.196 (1.007-4.786),respectively.Having found that Glasgow Coma Scale (GCS) is a strong predictor outcome(considering favourable outcome),ROC curve was made for finding out the optimal cutoff ( GCS=5) as a predictor.
Conclusion
The incidence of pulmonary infection in patients with severe head injury is high and prevention measures should be strengthened for high-risk patients to reduce the incidence of infection.

Clinical effect of different surgical methods for the treatment ofcommon bile duct stones

Objective
To compare the clinical effects of two surgical methods for the treatment of common bile duct stones.
Methods
The clinical data of 244 patients with common bile duct stones admitted to Binzhou Medical University Hospital in Shandong Province from January to September 2019 were retrospectively analyzed.All patients underwent surgery.Among them,144 patients underwent laparoscopic common bile duct stone removal (the experimental group),and 100 patients underwent conventional open choledocholithotomy (the control group).The intraoperative and postoperative complications of the two groups were compared and analyzed.Univariate analysis was performed using logistic regression analysis,and the Spearman correlation coefficient was used to test the correlation between the experimental group and the control group and other laboratory data.
Results
Intraoperative blood loss,anal exhaust time,hospitalization time,operation time and incision infection were statistically significant between the two groups.Postoperative complications such as bi-liary fistula and abdominal infection were not statistically significant.
Conclusion
Laparoscopic common bile duct incision and stone removal compared with conventional open bile duct incision and stone removal for common bile duct stones can effectively shorten the operation time and reduce intraoperative blood loss and postoperative complications.It has the advantages of small trauma,short hospitalization days and less complications,and it is worthy of promotion in hospitals,especially in primary hospitals.

Effect of hysteroscopic surgery on relative indicators of endometrial receptivity

Objective
To analyze the effect of hysteroscopic endometrial minimally invasive surgery on relative indicators of endometrial receptivity.
Method
Twenty patients with infertility from the gynecological clinic of a hospital from January 2017 to January 2018 were selected to observe endometrial morphology under hysteroscopy,collect endometrial specimens,and perform hysteroscopic endometrial minimally invasive surgery during implantation window stage.Hysteroscopic endometrial minimally invasive surgery was performed again 1 and 2 months after the operation,and endometrial specimens were collected at the implantation window 3 months after the operation.The changes of EP biochemical indexes before the operation and 3 months after the operation were compared.By HE staining,the secretion of endometrium was observed under an optical microscope.Immunohistochemical Streptomyces avidin peroxidase connection (SP) was used to detect the expression levels of estrogen receptor (ER),progestrone receptor (PR),leukaemia inhibitory factor (LIF),and vascular endotheli-al growth factor (VEGF) in endometrium.The levels of serum estradiol and progestrone during implantation window stage were analyzed.Embryo transfer was performed again,the implantation rate and pregnancy rate were followed up.
Results
The levels of serum estradiol and progestrone after surgery were slightly higher than those before the surgery,but there was no statistically significant difference.The expression levels of ER,PR and VEGF in endometrium after surgery were statistically significantly higher than those before the surgery (

<0.01) .The implantation rate and clinical pregnancy rate of IVF-ET were 45% and 35%,respectively.
Conclusion
Hysteroscopic endometrial minimally invasive surgery can significantly improve endometrial receptivity,the expression levels of ER,PR and VEGF in endometrium during implantation window stage,the implantation rate and pregnancy rate of IVF-ET.

Investigation of conjunctival sac bacteria and drug susceptibility in perioperative of cataract

Objective
To study conjunctival sac bacteria and drug susceptibility in perioperative of cataract.
Methods
Totally 264 cataract patients were selected and sampled at four time points after admission,before surgical disinfection,after surgical disinfection,and after surgery.Bacterial culture and drug sensitivity tests were conducted to compare the bacterial culture and drug sensitivity results in different periods.
Results
The positive rate of bacterial culture of conjunctival sac after admission was significantly higher than that before surgical disinfection,after surgical disinfection,and after surgery.After admission,the bacterial culture of conjunctival sac was mainly staphylococcus epidermidis.Gram-positive cocci are more sensitive to cefoxitin,gatifloxacin and levofloxacin.
Conclusion
Staphylococcus epidermidis is the most common bacteria in conjunctival sac.Antibiotics should be routinely applied before the operation,and providone iodine should be used to wash conjunctival sac to prevent posto-perative infectious endophthalmitis.

The effects of desflurane and propofol on inflammatory cytokinesand oxygen index in patients with one-lung ventilation

Objective
To summarize and analyze the effects of desflurane and propofol on serum inflammatory factors and o-xygenation index in patients with one-lung ventilation.
Methods
Patients undergoing elective thoracoscopic lobectomy in our hospital from November 2018 to May 2019 were randomly divided into two groups:group A (the desflurane group) and group B (the propofol group).The anesthesia methods were the same except the maintenance of anesthesia.The changes of patients'vital signs were monitored throughout the whole process.Arterial blood samples were collected at different time points for arterial blood gas detection,and the oxygenation index of patients was calculated.At the same time,the levels of IL-6,IL-8 and IL-10 in serum samples were detected.
Results
There was no significant difference in sex,age,weight,height and operation time between the two groups (

>0.05),and there was no significant difference in IL-6,IL-8,IL-10 and oxygenation index between the two groups (

Conclusion
There is no significant difference in the effects of desflurane and propofol on serum inflammatory factors and oxygenation index in patients with one-lung ventilation.Desflurane has a certain lung protective effect.

Study of alanyl-glutamine supplements on treatment compliance in elderly patientswith esophageal cancer on concurrent chemo-radiotherapy

Objective
To investigate the effect of alanyl-glutamine supplements in elderly patients with esophageal cancer on concurrent chemo-radiotherapy.
Methods
From November 2015 to November 2018,60 Elderly patients who were treated in our hospital with esophageal cancer on concurrent chemo-radiotherapy were recruited and randomly divided into the experimental group and the control group by random digits table.Both groups received routine nutritional support and dietary guidance,while only the experimental group received alanyl-glutamine supplements (20 g/d) on radiotherapy day.We compared blood indicators,changes in body weight,incidence of complications,completion rates of therapy and the T lymphocyte percentage of peri-pheral blood between the two groups.
Results
After chemoradiotherapy,albumin and hemoglobin of the control group decreased significantly,and the difference between the two groups was statistically significant,

<0.05.Compared with before treatment,the serum prealbumin level of the experimental group was higher than that of the control group,
P
<0.05.After chemoradiotherapy,serum transferrin of the control group decreased,

<0.05,and serum transferrin of the experimental group was higher than that of the control group,

<0.05.The incidence of radiation esophagitis and bone marrow suppression of the experimental group was significantly lower than that of the control group,

<0.05.There were no significant changes of body weight and incidence of radiation pneumonia before and after treatment in the two groups.The difference of immune indexes between the two groups was statistically significant,

Conclusion
For patients undergoing concurrent chemoradiotherapy for esophageal cancer,supplementing alanyl glutamine is beneficial to improve nutritional status,reduce the incidence of complications,reduce the possibility of immune decline,and thus improve the therapeutic compliance of patients.

Correlation between carotid atherosclerosis and ischemic cerebrovascular disease and the analysis of risk factors

Objective
To explore the correlation between carotid atherosclerosis and ischemic cerebrovascular disease and analyze the risk factors.
Methods
Eighty hospitalized patients with ischemic cerebrovascular disease and 80 healthy people with check-up were retrospectively analyzed,and were taken as the observation group and the control group respectively.The records were set down in the two groups of urinary inhibition C (CysC),serum triglyceride (TG),serum total cholesterol (TC),high-density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol (LDL-C) levels,the carotid artery diameter on both sides and film thickness (IMT) in the carotid artery.SPSS19.0 was used for statistical analysis and logistic regression analysis was carried out on the above factors.
Results
The differences of CysC,TC,HDL-C and IMT values between the two groups were statistically significant (
P
<0.05).Logistic regression analysis showed that CysC,HDL-C and bilateral IMT were correlated with ischemic cerebrovascular disease.The levels of CysC and bilateral carotid ARTERY IMT in the observation group were higher than those in the control group,and hdL-C level was lower than that in the control group (

Conclusion
Carotid internal-media thickness (IMT) is associated with ischemic cerebrovascular disease,while the increase of CysC and the decrease of HDL-C are the risk factors.

Clinical observation of method of invigorating spleen and regulating liver combined with metformin in the treatmentof 72 cases of type 2 diabetes mellitus with non-alcoholic fatty liver due to liver depression and spleen deficiency

Objective
To investigate the clinical effect of Tangyiping combined with metformin on type 2 diabetes mellitus with non-alcoholic fatty liver disease.
Methods
Seventy-two patients with type 2 diabetes mellitus and non-alcoholic fatty liver disease were randomly divided into the treatment group and the control group,with 36 patients in each group.The treatment group was given oral administration of Tangyiping combined with metformin,while the control group was given oral administration of metformin tablets for a course of 3 months.After the treatment,TCM syndrome score,blood lipid,blood glucose level and li-ver function were compared between the two groups.
Results
After 12 weeks of treatment,TCM syndrome,2 h PG,TC,HbAlc,and TG of the experiment group were significantly lower than those of the control group (

Conclusion
Tangyiping combined with metformin can improve TCM syndromes in patients with type 2 diabetes mellitus with liver depression and spleen deficiency combined with non-alcoholic fatty liver disease,reduce blood glucose and lipids,and improve liver injury and steatosis,so the effect is superior to that of metformin alone.

Promotion effect of concentrated growth factor extracton on the proliferation and differentiationof gingiva-derived mesenchymal stem cells

Objective
To evaluate the effect of concentrated growth factors extract (CGFe) on the proliferation of gingiva-derived mesenchymal stem cells (GMSCs) and differentiation of F-actin cytoskeleton.
Method
GMSCs were cultured in vitro.Cells in the experimental group were cultured with α-MEM containing CGFe (10% FBS),while cells in the control group were cultured with α-MEM without CGFe (10% FBS).The proliferation of GMSCs cells was determined by MTT assay,the activity of alkaline phosphatase (ALP) was detected at 1,3,5 and 7 d after osteogenesis induction,the formation of calcium nodules was detected by alizarin red staining at 14 and 21 d,and the morphology of the F- actin cytoskeleton was observed by confocal laser scanning microscope (CLSM).
Results
A significant increase of absorbance at 1,3 and 5 d was showed in the experiment group compared to that in the control group,

<0.05.On the assessment of ALP activity of GMSCs,the absorbance of the experiment group at 1,3 ,5 and 7 d was significantly higher than that of the control group,

<0.05 or

<0.01.On the assessment of osteogenic differentiation activity,the mineralized nodules were detected by the alizarin red (Sigma) staining.The overall absorbance of the calcium nodules in the experimental group at 7,14 d was significantly higher than that in the control group,

Conclusion
CGFe can significantly promote the proliferation of GMSCsand osteogenic differentiation.

Health economics evaluation of nosocomial infection in patients with COPD

Objective
To evaluate the economic loss caused by nosocomial infection in hospitalized patients with chronic obstructive pulmonary disease.
Methods
Totally 118 inpatients of COPD with nosocomial infection were selected as the case group,and according to the 1∶1 ratio,non-infection cases were taken as the control group.The basic information and effects of COPD on the hospital stay and hospitalization expense were compared between the two groups.
Results
The length of stay and the cost of hospitalization of patients with nosocomial infection of COPD increased,and the difference was statistically significant(
P<
0.01).There was significant difference between the two groups in the cost of each sub item except other costs(
P
<0.01).
Conclusion
Hospital infection of COPD patients prolongs the length of stay and increases the cost of hospitalization.Taking effective measures to control hospital infection will produce good economic and social benefits.

Effects of nursing intervention based on CITF on nursing satisfaction and self-care abilityof patients with hypertensive cerebral hemorrhage

Objective
To explore the effect of nursing intervention based on chronic illness trajectory framework (CITF) on nursing satisfaction and self-care ability of patients with hypertensive cerebral hemorrhage.
Methods
Patients with hypertensive cerebral hemorrhage were selected.The control group (62 cases) received conventional specialist nursing care,and the observation group (63 cases) received nursing prescriptions for chronic disease stage based on conventional care.Nursing satisfaction scale (NSNS) and self-care ability scale (ESCA) were used to compare the differences in nursing satisfaction and self-care ability scores over time before and after the two interventions.
Results
The scores of nursing satisfaction and self-care ability with time nodes in the observation group were higher than those in the control group (
P
<0.05).
Conclusion
Nursing intervention based on CITF framework can better meet the different needs of patients,improve nursing satisfaction and self-care ability of patients,and promote patient rehabilitation.
