Expression of microRNA-212 in cervical cancer and its effect on proliferation and invasion of cancer cells

Objective
To investigate the expression of miR-212 in cervical cancer and its effect on proliferation and invasion of cervical cancer cells and its mechanism.
Methods
RT-PCR and Western Blot were used To detect the expression levels of miR-212 and TCF7L2 in cervical cancer cells. BrdU cell proliferation assay and Transwell invasion assay were used to detect cell proliferation and invasion. Target Scan was used to predict the protein-coding gene target of miR-212, and double luciferase reporting system was used to validate it.
Results
The expression of mir-212 in cervical cancer tissues was down-regulated, compared with that in adjacent tissues (
P
<0.01). Compared with End1/E6E7 cells, the expression level of mir-212 in cervical cancer cell lines was significantly reduced (
P
<0.01). Overexpression of miR-212 can inhibit cell proliferation and invasion, while low expression of miR-212 can promote the proliferation and invasion of cervical cancer cell lines (
P
<0.01). Overexpression of miR-212 can significantly inhibit the expression of TCF7L2 protein, while low expression of miR-212 can promote the expression of TCF7L2 protein. The target gene TCF7L2 is the direct target of miR-212.
Conclusion
The expression of miR-212 in cervical cancer tissue was decreased, and the invasion and metastasis of cervical cancer were inhibited by targeting regulation of TCF7L2 gene.

The significance of DNA quantitative analysis combined with hMSH6 in cervical cancer screening

Objective
To investigate the correlation and clinical significance of DNA ploidy analysis with hMSH6 and HPV in cervical cancer.
Methods
High risk HPV-DNA detection and immunohistochemical hMSH6 staining were used to analyze the expression of hMSH6, HPV infection and correlation in cervical tissues with different lesions.
Results
The specificity of cervical lesions was 25.86% and 77.59%, respectively, based on the presence of DNA aneuploidy and moderate amount of DNA aneuploidy. It suggests that the presence of more than moderate amount of DNA aneuploidy has a higher diagnostic value for cervical lesions screening. The positive rates of hMSH6 were 75% and 85.71% in the multidose and medium dose alloploid groups, which had statistically significant differences from the control group (
P
<0.05). The infection rate of HPV was 92.86% and 76.19% in the multi-dose and medium-dose DNA aneuploidy groups,which showing statistically significant differences from the control group (
P
<0.05). The MSH6 positive rate of the multiploidy abnormal HPV infection group was 90.38%, which was different from the MSH6 positive rate of the multiploidy abnormal HPV infection group 25% (
P

Conclusion
HPV combined with hMSH6 test is the detection of etiology, with high sensitivity. The high expression of hMSH6 protein caused by HPV infection is expected to provide a reference for the prevention and treatment of cervical cancer.

Change of liver function and regeneration after portal vein ligation dombined with in situ splitting on in cirrhosis rats

Objective
To test serum levels of ALT,AST,ICG R15 and calculating the hepatic regeneration rate (HRR) of remnants (median lobe) on normal and cirrhosis rats,to explore the rule of the liver function and regeneration after operation and analyse the correlation.
Methods
Thirty male SD rats with normal and cirrhosis (induced by CCl
4
) were randomly selected. The two group rats were divided into 5 subgroups, including the 1st, 3rd, seveth, tenth, and fourteenth day subgroups. All of the rats were underwent ALPPS and were tested AST、ALT and ICG R15, and regeneration rate of residual liver was count.
Results
AST, ALT of two groups were raised after surgery, the experimental group was obviously higher than the control group, which had statistical difference. Next to the two levels were decreased gradually, the control group was normal in 7 days after surgery, while the experimental group was normal in 14 days after surgery. ICG R15 had the same change. Regeneration rate of residual liver increase gradually, but regeneration rate of the control group was faster than the experimental group. The correlation analysis showed thatthey had obvious correlation (
r
=0.841). The single factor linear regression analysis obtained an equation of regression: the control group was Y= -0.1647x+39.396 (X was ICG R1 5, Y was regeneration rate of residual live).
Conclusion
The liver function injury of the experimental group was more obvious than the control group, and the recovery of liver function was slower than the control group, too. And the liver function had correlation with liver regeneration. So the volumes of residual live could be used as an index for evaluating liver function.

Short-term analysis of decompression with or without fusion treatment for degenerative lumbar spondylolisthesis

Objective
To compare the short term effects of decompression with or without fusion in the treatment of degenerative lumbar spondylolisthesis.
Methods
A total of 105 patients underwent decompression with or without fusion. According to the inclusion and exclusion criteria, totally 78 patients were enrolled, including 52 patients with decompression without fusion and 26 patients with fusion. The data were collected and recorded including age, sex, hospital stay, operative blood loss, operation time, bedtime, bone fusion, preoperative and postoperative10 days, 3 months, last follow up VAS, preoperative and postoperative 3 months, last follow up ODI. The follow up time was more than 12 months.
Results
The operative blood loss, operation time, hospital stay, bedtime, complications, postoperative10 days VAS of decompression with fusion group were significantly higher than those without fusion (
P
<0.05). The VAS and ODI scores of the postoperative 3 months, last follow up were similar (
P
>0.05).
Conclusion
Decompression with or without fusion are safe and effective surgical methods for the treatment of degenerative lumbar spondylolisthesis. Decompression without fusion has less surgical trauma, less bleeding, less complications and less cost.

Analysisi of the influence of femoral prosthesis rotation angle and tibia varus angle after tibial osteotomy on the mechanical axis alignment after computer-navigated assisted TKA

Objective
To simulate total knee arthroplasty under the monitoring of the computer navigation system, and to observe the effects of different femoral prosthesis rotation angle and tibia varus angle on the mechanical axis alignment.
Methods
Total knee arthroplasty was performed on synthetic lower limb using computer navigation system. Mechanical axes at 0°,30°,60°and 90° were calculated when 0°,3°and 5° tibia varus under different femoral prosthesis malrotation, respectively, and the change trend and rule were obtained.
Results
Under the condition of insufficient external rotation of femoral prosthesis, the mechanical axis varus angle of lower limbs decreased with the increase of varus angle after tibial osteotomy(
P
<0.05 or <0.01). In the case of insufficient external rotation of the femoral prosthesis, the varus angle of the lower limb gradually decreased within the interval of 0°~90° for the same posterior varus angle of the tibial osteotomy(
P
<0.05). In the case of excessive external rotation of the femoral prosthesis, the varus angle of the lower limb increased with the gradual increase of the varus angle after the tibial osteotomy(
P
<0.05 or <0.01). In the case of excessive external rotation of the femoral prosthesis, the varus angle of the lower extremity gradually increased within 0°~90° at the same posterior varus angle of the tibial osteotomy(
P
<0.05).
Conclusion
Rotational alignment for femoral prosthesis and tibia varus were important factors in the achievement of accurate mechanical axis alignment.

Meta-analysis of short-term efficacy of robotic and laparoscopic gastrectomy for gastric cancer

Objective
To compare the feasibility and short-term efficacy of robotic and laparoscopic gastrectomy for gastric cancer.
Methods
The literatures about the short-term efficacy of robotic gastrectomy (RG) and Laparoscopic gastrectomy (LG) from gastric cancer were collected. There are 849 patients receiving TG and 1 141 patients receiving LG were delecteded in the literature. The extrate data about operation time, blood loss, lymph node dissection, restore fluid diet time, first exhaust time after surgery, hospital stay after surgery, and short-term complications after surgery from the two groups were needed. This meta-analysis use RevMan 5.3 software to analyze data and we divided the studies into three groups for subgroup analysis.
Results
A total of 150 documents and 14 were selected. Among them, 849 patients with gastric cancer received TG, and 1 141 patients received LG. The results showed that there were significant differences in the operation time, blood loss, number of lymph nodes removed, first exhaust time after surgery, and hospital stay in the RG group and the LG group (
P
<0.05). There was no significant difference in restore fluid diet time and short-term complications
(P
>0.05).
Conclusion
Compared with LG, RG was a safe and feasible treatment for patients with gastric cancer. However, RG is not completely better than LG.From now on. It should focus on decreasing the operation time and reducing the incidence of complications after surgery to improve the effectiveness of treatment .

Application of chromosome microarray analysis technology in prenatal diagnosis

Objective
To investigate the clinical application value of chromosomal microarray analysis (CMA) in prenatal diagnosis.
Methods
Totally 132 pregnant women who underwent amniocentesis in Binzhou Medical University Hospital from July 2018 to June 2019 were selected as subjects. The traditional karyotype analysis and CMA test were performed in all cases. The data acquired from these two methods were statistically analyzed.
Results
Thirteen cases with chromosomal aneuploidies, 1 case with unbalanced rearrangements were detected both from karyotyping and CMA, while CMA could more accurately give the abnormal sites of copy number variation. Submicroscopic rearrangements with pathological significance were detected in 3 cases with normal karyotypes (+2.27%) and CNV of unclear significance were detected in 4 cases (3.03%). However, the chromosome inversion, polymorphism and balance translocation could not be detected by CMA.
Conclusion
Compared to karyotype analysis, CMA technology has the same detection efficiency for aneuploidy and copy number variations, while CMA has higher sensitivity and resolution, and can also find additional clinical significant gene copy number. However, the CMA analysis technique itself has limitations, so CMA cannot completely replace the traditional karyotype analysis technology.

Clinical effect of laparoscopy on early endometrial cancer

Objective
To observe the clinical effect of laparoscopy on early endometrial cancer.
Methods
A retrospective analysis was conducted. Sixty patients with early endometrial cancer who underwent laparoscopic surgery in our hospital from February 2016 to March 2018 was the observation group, and 60 patients who underwent laparoscopic surgery during the same period were the control group. Perioperative indicators were compared between the two groups such as operation time, intraoperative blood loss, postoperative hospitalization time, postoperative exhaust time, and urinary indwelling time, and the incidence of postoperative complications, postoperative recurrence rate, and tumor-free survival rate were recorded in the two groups.
Results
The intraoperative blood loss, postperative hospitalization in the observation group were significantly better than those in the control group (
P
<0.05). There was no significant difference in the number of lymphatic clearance between the two groups (
P
>0.05). The postoperative complications rate in the observation group was significantly lower than that in the control group (
P
<0.05). Compared with the control group, the recurrence rate, tumor-free survival rate, and mortality index within the observation group within 3 years were not significantly different (
P
>0.05).
Conclusion
Laparoscopic surgery is ideal for treatment, with minimal trauma, high safety, and rapid recovery after surgery, and is worthy of clinical promotion.

Retrospective analysis of pregnancy risk in the advanced maternal age

Objective
To provide a scientific basis for strengthening the health care of high-age women during pregnancy and improving pregnancy outcome.
Methods
Totally 2 994 cases in the advanced maternal age group≥35 years and 16 172 cases in child-bearing age pregnancy at the same time of inpatient delivery were collected, and the medical records of the two groups were compared and analyzed.
Results
The complications of gestational diabetes mellitus, preplacental placenta and placental abruption in the advanced maternal age group were significantly higher than those in the child-bearing age group (
P
<0.05), and the incidences of adverse outcomes such as fetal growth restriction and fetal malformation in the advanced maternal age group was significantly higher than those in the child-bearing age group (
P
<0.01). The incidence of intervention preterm delivery in the advanced maternal age group was higher than that in the child-bearing age group (
P<
0.01).
Conclusion
In order to reduce the incidence of comorbidities and long-term complications, to ensure the safety and health of mothers and infants, we should promote appropriate age pregnancy.

Effect evaluation of the peer education model on health education in patients with rheumatoid arthritis

Objective
To investigate the effect of peer education mode on education in RA patients.
Methods
By using the random number table method, 160 RA patients discharged from a tertiary hospital in Binzhou were selected and divided into the control group 80 cases and the intervention group 80 cases. The control group adopted the regular telephone follow-up health education intervention mode, while the intervention group adopted the peer education mode, and the intervention period was 3 months. The quality of life (SF-36 scale), anxiety and depression (SAS and SDS scale) and medication compliance (Morisky medication compliance scale) of the two groups before and after the intervention were compared.
Results
The scores of energy VT, social function SF, emotional function RE and mental health MH in the intervention group were higher than those in the control group (
P
<0.05); the SAS and SDS scores of the intervention group were higher than those of the control group (
P
<0.05). Three months after discharge, The medication compliance of the intervention group was better than that of the control group (
P
<0.05).
Conclusion
Peer education can effectively improve patients'anxiety and depression, promote their active cooperation, improve their medication compliance, and further improve their quality of life.

Expression of sclerostin in alveolar bone of experimental periodontitis in rats

Objective
To observe the expression and distribution of sclerostin in alveolar bone of experimental periodontitis in rats.
Methods
16 SD rats of about 3 months old were randomly divided into 4 groups (1 blank control group and 3 experimental groups). The rats were sacrificed at 0, 3, 7, and 14 d, and the experimental tissue was taken. The expression and distribution of sclerostin were detected by immunohistochemistry.
Results
The model of rat periodontitis is made by ligating the teeth with silk, and the method is mature and reliable. Sclerostin was occasionally expressed in alveolar bone tissue of normal adult rats. On the third day, the number of sclerostin-positive bone cells increased exponentially, the 7 d group reached a peak, and the 14 d group began to decrease.
Conclusion
The cells participate in the pathogenesis and development of periodontitis through the synthesis of sclerostin; the destruction and reconstruction of alveolar bone, the expression shows a certain regularity with time.

Detection effect of gingival crevicular fluid and serum matrix metalloproteinases in pregnant women with periodontitis on preterm birth prediction

Objective
To evaluate the predictive value of matrix metalloproteinases in preterm labor in pregnant women with periodontitis and examine the hypothesis that periodontitis was one of the sources of maternal inflammation in premature infants.
Methods
The samples of gingival crevicular fluid (GCF) and serum were collected from groups of 62 periodontitis mothers (PG) and 60 oral healthy mothers (HG). Levels of MMP-9 and MMP-25 were analyzed by enzyme-linked immunosorbent assay (ELISA).
Results
The gestational weeks of pregnant women in the PG were significantly fewer than those in the HG (
P
<0.05). MMP-9 and MMP-25 expression levels in GCF and serum were significantly higher in the PG than in the HG (
P
<0.05).
Conclusion
There was a tendency presenting the higher expression of MMP-9 and MMP-25 in CGF and serum, the shorter gestational period. Detection of the expression level of MMP-9 and MMP-25 in the body fluids of pregnant women with periodontitis in middle and late pregnancy can be used as a target to reduce the preterm rate.

Methods

Bge., quantitative PCR was used to detect the mRNA expression levels of inflammatory factors iNOS, IL-1β, and IL-6.
Results
The purity of total flavonoids of
Potentilladiscolor
Bge. obtained by polyamide chromatography was 87.4%, and the yield was 82.6%. The total flavones of
Potentilladiscolor
Bge. could significantly down-regulate the mRNA expression of inflammatory factors iNOS, IL-1β and IL-6 in RAW 264.7 cells induced by LPS.
Conclusion
The total flavonoids of
Potentilladiscolor
Bge. can be prepared by polyamide resin chromatography, and its anti-inflammatory activity is related to the down-regulation of mRNA expression of iNOS, IL-1β, and IL-6.

Synthesis and anti-inflammatory activity of resveratrol derivatives

Objective
To obtain better drug lead compounds with anti-inflammatory activity from derivatives of natural product resveratrol.
Methods
Resveratrol was used as a lead compound for structural modification and 6 derivatives were designed and synthesized, including one methylation product, four esterification products and one amide analogue.The structures of all derivatives were characterized by 1H-NMR, and MS spectra. The inhibition on NO production of resveratrol derivatives in LPS-stimulated RAW 264.7 cells were tested by Griess method.
Results
The results indicated that amide derivative 6 possessed some application potential with good anti-inflammatory activities and no cytotoxity.
Conclusion
Research on the potential anti-inflammatory activity of resveratrol derivatives can provide a material basis for the research of new anti-inflammatory drug lead compounds.

Diagnostic value of lung ultrasound with high-resolution CT for interstitial lung disease

Objective
To compare the sensitivity of lung ultrasound (LUS) with high-resolution computed tomography (HRCT) in the diagnosis of interstitial lung disease (ILD).
Methods
A retrospective analysis of 30 patients with clinically diagnosed ILD who were hospitalized in Binzhou Medical University Hospital from June 2018 to June 2019. All patients underwent bilateral LUS examination and chest high-resolution CT (HRCT) scan with informed consent. The interval between inspection and HRCT is no more than 24 hours.Record the pleural and lung parenchymal sonograms, observe the number of B lines, the thickness of the pleural line, whether the pleural line is regular, whether there is a hypoechoic area under the pleura, and whether there is fluid in the chest. The sensitivity of LUS and HRCT in the diagnosis of ILD was calculated by clinical diagnosis as the gold standard.
Results
Of the 30 patients with ILD, 4 patients (13.3%) had false negative for LUS and 3 (10%) had false negative for HRCT. No false positive cases were found. All patients were ILD, so the specificity of the examination could not be counted. With the clinical diagnosis of ILD as the gold standard, the sensitivity of LUS and HRCT for the diagnosis of ILD was 0.87 (0.69-0.96) and 0.90 (0.73-0.98), respectively. There was no significant difference in sensitivity between the two groups (

=1.00).
Conclusion
LUS is not inferior to HRCT in the diagnosis of ILD. Considering its safety, low cost, portability and practicability, LUS can be used as a potential imaging tool to replace HRCT screening ILD.

Research progress on the interactions between homocysteine and epilepsy

Homocysteine is formed as an intermediate during methionine metabolism. Hcy is known to be an independent risk factor for cardiovascular disease and neurological diseases including epilepsy, stroke, cognitive disorders, Parkinson's disease and Alzheimer's disease. Epilepsy is one of the common diseases of neurology, and it has characteristics of episodic, transient, repetitive and stereotypic. One research has shown that severe hyperhomocysteinemia causes seizures and increases the frequency of seizures. But the interactions between hyperhomocysteinemia and seizures are rare reported in China. This review focuses on the interactions among homocysteinemia, NMDA receptors and epilepsy to further explore the pathogenesis of epilepsy and provides new clinical treatments.
