Effect of IL-17A on the expression of gelatinase in osteoblasts MC3T3-E1 cells and its mechanism

Objective
To investigate the effect of IL-17A on gelatinase expression in MC3T3-E1 cells and to explore its mechanism.
Methods
MC3T3-E1 cells were stimulated by IL-17A. The expression of MMP-2 and MMP-9 at mRNA and protein levels were detected by Real-time PCR and ELISA respectively. Immunofluorescence and Western Blot were used to detect the IL-17A-induced activity of NF-κB. Chemical inhibitor of NF-κB was used to detect the involvement of NF-κB in IL-17A-induced MMP-9 expression in MC3T3-E1 cells.
Results
IL-17A upregulated MMP-9 expression at both mRNA and protein levels (

<0.01), but had no effect on MMP-2 expression; IL-17A promoted phosphorylation of NF-κB (
P
<0.01); inhibition of NF-κB attenuated IL-17A-induced MMP-9 expression(

<0.05). IL-17A did not affect the TIMP-1 and TIMP-2 expression at mRNA level.
Conclusion
IL-17A can promote the secretion of MMP-9 by MC3T3-E1 cells by activating NF-κB.

Preparation of acellular matrix facial nerve repair material

Objective
To prepare the acellular matrix facial nerve repair material and construct a tissue-engineered facial nerve with a natural neural tissue structure which can be used for the repair of peripheral nerve injury.
Methods
The rabbit facial nerve was taken, and acellular nerve matrix was prepared by chemical extraction, the degree of decellation and demyelination was observed by HE staining.The DNA content of the cells in the scaffold after staining with Quant-iT PicoGreen working solution were measured by fluorescence spectrophotometer. The relative growth rate of cells on the scaffold was determined by MTT to detect the cytotoxicity.
Results
The scaffold graft was cylindrical, and its elasticity was basically consistent with normal nerves. Tissue observation showed that the structure of cell, the nerve myelin sheath and axon had been completely removed. The extracellular matrix formed a longitudinal arrangement structure and was filled with voids. The residual DNA content in the rabbit acellular facial nerve matrix scaffold was significantly lower than that in the normal rabbit facial nerve. Nerve matrix donors were non-cytotoxic.
Conclusion
The modified chemical extraction method can effectively remove facial nerve cells, preserve the natural structure intact and have low cytotoxicity. It can be used as a scaffold for tissue engineering facial nerves.

Regulatory effects of human placenta derived mesenchymal stem cells on the treatment for OA-induced acute lung injury

Objective
To investigate the therapeutic effect of human placenta mesenchymal stem cells (hPMSCs) on OA-induced acute lung injury and its possible mechanism.
Methods
Thirty-six SD rats were divided into the control group, the lung injury group and the treatment group. Acute lung injury rat model was prepared by sublingual intravenous injection of oleic acid. After the successful modeling, the samples were kept for 24 h after the corresponding treatment, and the lung tissue morphology, the ratio of wet to dry mass of lung tissue (W/D), the proportion of neutrophils in alveolar lavage fluid and the serum IL-10, IL-8 level were detected.
Results
Pulmonary histopathology showed that normal alveolar structure was normal in the normal group, typical ALI changes were observed in the lung tissue of the lung injury group, and the degree of injury in the cell transplantation group was relatively mild. Compared with the control group, the number of neutrophils in bronchoalveolar lavage fluid was significantly increased in the injury group and the transplantation group (
P<
0.05 or <0.01), and the wet/dry ratio of bronchoalveolar lavage solution was also increased in the injury group and the transplantation group (
P
<0.05). Compared with the lung injury group, the number of neutrophils and the wet/dry ratio of bronchoalveolar lavage fluid in the transplantation group were all decreased (

<0.05). Plasma and lung tissue levels of IL-8 and IL-10 in the injury group and the transplantation group were significantly increased, compared with the control group (

0.05 or <0.01); IL-8 levels in the transplant group were significantly reduced compared with the injury group (

<0.05 or <0.01).
Conclusion
Placental mesenchymal stem cell transplantation in the treatment of acute lung injury can reduce the level of inflammatory factors, improve the level of anti-inflammatory factors, stabilize the internal environment, avoid the occurrence of inflammatory storms, and provide an effective treatment method for the clinical treatment of critical patients.

Application of intra-voxel incoherent motion combined with biomarker in the evaluation of efficacy of non-small cell lung cancer

Objective
To investigate the role of IVIM parameters and serum VEGF and HIF-1α in evaluating the efficacy of chemoradiotherapy for advanced non-small cell lung cancer (NSCLC).
Methods
Fifty-nine cases of advanced NSCLC were treated with chemoradiotherapy. IVIM was performed before and after treatment, and the parameters before and after treatment were obtained by post-treatment software,serum VEGF and HIF-1α levels were obtained before and after treatment.Response to chemoradiotherapy was assessed as complete response (CR), partial response (PR), disease stability (SD) and disease progression (PD) according to RECIST 1.1. CR and PR were classified as curative effect sensitive group.SD and PD were classified as efficacy insensitivity group.The independent sample T test was used to compare the changes of each parameter value before and after treatment between the two groups, and the ROC curve was used to analyze each parameter to judge the evaluation efficacy of concurrent chemoradiotherapy for advanced NSCLC, and the optimal threshold was determined.
Results
Serum VEGF and HIF-1α in NSCLC patients were significantly higher than those in healthy volunteers (

<0.01).IVIM parameters and serum VEGF and HIF-1α levels were correlated with age, stage and pathological type (

VEGF and HIF-1α alone or in combination with IVIM parameters can be used as effective indicators for early prediction of NSCLC chemoradiotherapy efficacy.

NEAT1 expression in gastric cancer and its correlation with clinicopathological parameters of patients with gastric cancer

Objective
To study the clinical significance of LncRNA NEAT1 in gastric cancer.
Methods
The expression level of LncRNA NEAT1 in gastric cancer tissues and adjacent normal tissues was detected by fluorescence quantitative PCR, and the correlation between the expression level of NEAT1 and the clinicopathological parameters and prognosis of gastric cancer patients was further analyzed.
Results
The expression of NEAT1 in gastric cancer tissues was higher than that in normal tissues (
P<
0.01). The expression of NEAT1 was significantly correlated with vascular invasion, distant metastasis, and recurrence in patients with gastric cancer (
P
<0.05), but not with age, gender, degree of differentiation, nerve invasion, staging, and lymph node metastasis. Univariate and multivariate COX regression analysis showed that the expression level of NEAT1 was related to disease-free survival and overall survival of patients, and could be used as an independent risk factor for the prognosis of gastric cancer patients. Survival analysis showed that gastric cancer patients in the NEAT1 group with high expression had a poor prognosis.
Conclusion
NEAT1 may play a role in promoting cancer in gastric cancer, and can be used as a potential indicator for clinical diagnosis and prognosis evaluation.

Meta-analysis of effectiveness and safety on large-artery atherosclerosis-induced cerebral infarction with combination therapy of argatroban and clopidogrel

Objective
To systematically analyze the effectiveness and safety on large-artery atherosclerosis-induced cerebral infarction with combination therapy of argatroban and clopidogrel.
Methods
Literatures about ischemic cerebral infarction with combination therapy of argatroban and clopidogrel were searched in the databases of CNKI, VIP, Wanfang, CBM, Pubmed, Cochrane Library, and Embase from the beginning of the databases to October lst, 2019. Data extraction and quality evaluation were performed after a RCT that met inclusion and exclusion criteria. Meta-analysis was performed by Review Manager 5.3.
Results
Seven RCTs and 678 subjects were included in our analysis. The results of meta-analysis suggested that the combination therapy group was better than the control group in improving patients' therapeutic efficiency (

<0.001), NIHSS scores (
MD=-1.97, P
<0.001), and Barthel scores (
MD=7.85, P<
0.001). In addition, no significant association was found between two groups in bleeding.
Conclusion
The current evidence indicates that combination therapy of argatroban and clopidogrel could improve therapy efficiency, neurological impairment and activities of daily living abilities in patients with large-artery atherosclerosis-induced cerebral infarction.

The clinical efficacy of bevacizumab combined with pemetrexed in the maintenance treatment of advanced lung adenocarcinoma

Objective
To observe the clinical efficacy and side effects of pemetrexed combined with bevacizumab in the maintenance treatment of advanced lung adenocarcinoma.
Methods
Fifty-four patients with driver gene negative advanced lung adenocarcinoma received 4-6 cycles of first-line pemetrexed +/- bevacizumab + cisplatin chemotherapy. After the disease was controlled, they were divided into the pemetrexed + bevacizumab + cisplatin group and pemetrexed monotherapy group, and then the PFS, effective rate and adverse reactions of the two groups were observed. At the same time, fasting venous blood of patients before maintenance treatment and after 4 cycles of maintenance treatment was collected, and the vascular endothelial growth factor (VEGF) level was detected by ELISA to evaluate the correlation between changes in serum VEGF level and the efficacy.
Results
Both ORR and DCR of the two drug groups were significantly higher than those of the single drug group (
P
<0.05 or <0.01). The median PFS of two groups were 7.8 and 3.9 months and there was statistically significant difference between the two groups by log-rank test. After 4 cycles of maintenance treatment, the serum VEGF level of patients in the two groups significantly decreased after treatment, compared with VEGF level before treatment (
P
<0.01); compared with the single drug group, the serum VEGF level of patients in the two drug groups decreased significantly after treatment (
P<
0.01).
Conclusion
The combination of pemetrexed and bevacizumab in the maintenance treatment of advanced lung adenocarcinoma is more effective in the near term, with better efficacy and safety, and is worthy of clinical promotion. Meanwhile, dynamic monitoring of serum VEGF expression level can effectively predict the efficacy of anti-tumor therapy and is convenient and non-invasive.

Correlation between bone density T value and coronary heart disease in the elderly

Objective
To investigate the correlation between bone mineral density T value and coronary heart disease in the elderly.
Methods
Retrospective analysis was performed on 512 elderly patients who underwent coronary angiography and bone mineral density examination.According to the results of coronary angiography, 273 patients in the coronary heart disease group and 239 patients in the non-coronary heart disease group. According to the results of DXA bone mineral density examination, there were 302 cases of low bone density group and 210 cases of normal bone density group. The general conditions of each group of patients, the difference of laboratory test indicators, and bone density T value were compared. Logistic regression analysis was used to analyze the risk factors of coronary heart disease in the elderly.
Results
In 512 elderly patients, the T-value of bone density was lower in the coronary heart disease group and the risk of low bone mineral density was higher in the non-coronary heart disease group (
P<
0.05 or<0.01). Compared with the normal bone density group, the group with low bone mineral density had the high proportion of older, female and diabetic patients, the T-value of bone mineral density was lower, and uric acid was higher (
P
<0.05 or<0.01). Correlation analysis showed that the bone density T value was negatively correlated with age, ALP, FBG (
P
<0.01), the bone density T value was positively correlated with uric acid (
P
<0.001). Regression analysis showed that aging, diabetes, were risk factors for low bone density. The results of single factor and multivariate logistic regression analysis showed that high bone density T value was an independent risk factor for coronary heart disease (

Conclusion
Low bone density T value is an independent risk factor for the onset of coronary heart disease in the elderly.

Survey on iodine nutritional status of pregnant women and thyroid function of neonates in Binzhou

Objective
To investigate the effects of different iodine nutrition status on thyroid function in pregnant women and TSH levels in newborns.
Methods
Two hundred and eighty-four pregnant women were selected in the third trimester of pregnancy (≥28 weeks of pregnancy) of a hospital in 2019.The morning-urine was collected to detect the levels of free triiodothyronine (FT

), free thyroxine (FT

) and sensitive thyroid stimulating hormone(sTSH).The venous blood samples were collected to detect the content of iodine in urine,chemiluminescencetthyroid peroxidase antibody (TPOAb) and thyro-globulin antibody (TGAb). Heel blood of 72 hours of normal term was collected to detect the levels of TSH.
Results
The ratios of urinary iodine in women in Binzhou area were below 149 μg/L, 150~249 μg/L, 250~499 μg/L, and above 500 μg/L were 18.31%, 13.73%, 19.01%, and 48.94%, respectively. There were 41 pregnant women suffering from thyroid disease (14.44%), 25 pregnant women suffering from methylene chloride (8.80%), and 16 pregnant women suffering from hypothyroidism (5.63%).The neonatal heel blood TSH level was (2.95 ± 2.50) mIU / L and 15.32% of neonatal heel blood TSH was higher than 5mIU / L. There was no significant difference in heel heel blood TSH of newborns born by pregnant women with different levels of iodine nutrition (
P
>0.05).
Conclusion
The iodine deficiency and iodine excess problems of pregnant women in Binzhou area coexist. The proportion of pregnant women with iodine excess is high, and the TSH level of newborns is high. The iodine nutrition status of pregnant women can affect the iodine nutrition level of newborns.

Effects of ketorolac tromethamine combined with propofol anesthesia on hemodynamics and stress response in elderly patients undergoing painless gastroscopy

Objective
To investigate the effects of ketorolac tromethamine combined with propofol on hemodynamics and stress response in elderly patients with painless gastroscopy.

The expression and correlation analysis of linc00673 in plasma of patients with colorectal cancer between clinical features

Objective
To detect the expression of linc00673 in patients with colorectal cancer, and then explore the relationship between the expression of plasma linc00673 and clinical symptoms.
Methods
Totally108 hospitalized patients with colon polyps and colorectal cancer admitted to Shenzhen People's Hospital were enrolled. Real-time PCR was used to detect the difference in the expression of linc00673 in plasma of colorectal cancer and rectal polyps. The correlation between linc00673 and patient age, gender, TNM tumor stage, tumor location, gross type and lymph node metastasis and tumor volume was analyzed.
Results
Linc00673 was highly expressed in colorectal cancer tissues(

<0.05). Correlation analysis showed that high expression of linc00673 was not associated with age, gender, TNM tumor stage, tumor location, gross type and lymph node metastasis, and was positively correlated with tumor volume (

Conclusion
The high expression of linc00673 in colorectal cancer tissues suggests that linc00673 may be a potential marker for the diagnosis of colorectal cancer, which may provide a theoretical basis for the diagnosis and targeted therapy of colorectal cancer.

Effects of gasbag midwifery assisted vaginal delivery on labor process and pregnancy outcomes of puerperae with scarred uterus and previous history of cesarean section

Objective
To analyze the effects of gasbag midwifery assisted vaginal delivery on labor process and pregnancy outcomes of puerperae with scarred uterus and previous history of cesarean section so as to provide a reference for clinical delivery of such women.
Methods
The control group underwent routine vaginal delivery, while the observation group underwent gasbag midwifery assisted vaginal delivery. The delivery mode, labor process and pregnancy outcomes were compared between the two groups.
Results
There was no significant difference in the delivery mode or pregnancy outcomes between the two groups (
P
>0.05). The first stage of labor, the second stage of labor and the total labor time of the observation group were significantly shorter than those of the control group (

0.05).
Conclusion
Gasbag midwifery assisted vaginal delivery can effectively shorten the labor process of puerperae with scarred uterus and previous history of cesarean section, without increasing the risks of conversion to cesarean section, forceps delivery or adverse pregnancy outcomes.

Role of behavior management in dental treatment of preschool children with tooth decay

Objective
To investigate the effect of behavioral management on dental anxiety during the diagnosis and treatment of dental caries in preschool children, and to evaluate its role in relieving dental anxiety and improving treatment behavior.
Methods
Totally 500 children met the inclusion criteria and were randomly divided into the control group

=250) and the experimental group (

250). Behavior management techniques were used in the experimental group and routine nursing methods were used in the control group. Children's fear survey schedule-dental subscale (CFSS-DS) and clinical cooperation rate were compared to evaluate the effect of behavioral management.
Results
The mean CFSS-DS score of children in the experimental group was significantly lower than that in the control group (

<0.05). The incidence of anxiety in the experimental group was lower than that in the control group (

<0.05). The clinical cooperation rate of children in the experimental group was higher than that in the control group (

Conclusion
The implementation of behavioral management techniques, such as cognitive behavioral therapy, speaking-demonstration-doing, systematic desensitization, distraction, voice control, positive reinforcement and exemplary role could significantly alleviate children's dental anxiety and effectively improve the rate of treatment cooperation.

Effect of different amount of exercise on intestinal preparation and cleaning in elderly patients

Objective
To study the effect of different amount of exercises on the cleaning effect of intestinal preparation in elderly patients.
Methods
This study collected 170 elderly patients who underwent colonoscopy in a tertiary hospital from May 2019 to October 2019. They were divided into 2 groups, A and B groups, each with 85 patients. Group A: For each 500 mL of PEG solution, the patient's exercise steps should be controlled at 300-500 steps, and the total exercise steps should be controlled at 2 000-3 000 steps; Group B: For each 500 mL of PEG solution, the patient's exercise steps should be controlled at 500-900 steps, and the total number of exercise steps was controlled between 3 000-5 000 steps; We observed and compared the total medication time, first defecation time, total defecation times, last defecation characteristics, and intestinal preparation quality of the two groups of patients.
Results
The qualified rate of intestinal preparation and the Boston intestinal preparation scale (BBPS) scores of group B were higher than those of group A (

<0.01). The total medication time and the first defecation time were shorter in group B. The number of defecations increased, and the last defecation traits were better than those in group A (

Conclusion
Controlling the total number of exercise steps at 3000-5000 steps can effectively shorten the total time for intestinal preparation and the first defecation time for elderly patients, and increase the total number of defecations after taking the medicine to obtain the ideal intestinal preparation effect.

Biosynthesis of ε-polylysine and its application in medicine field

ε-polylysine is a kind of natural amino acid polymer produced by microbial fermentation, which has a broad application prospect in food, medicine and other industries. This paper reviewed the research progress in the biosynthesis of ε-polylysine in recent years, and especially discusses the new breeding methods of ε-polylysine production strains and the application of ε-polylysine in medicine field. The aim is to provide theoretical basis for the industrial production and application development of ε-polylysine.

Advanced progress of Th17 cell in malignant tumors

T helper cell 17 (Th17) is a specific subset of T helper lymphocytes determined by high secretion of IL-17 and other inflammatory cytokines. Compared with the typical Th1/Th2 cells, Th17 cells show stronger instability and plasticity, and play a part in different diseases, including infectious diseases, autoimmune disorders and cancer. Th17 cells increase tumor progression by activating angiogenesis and immunosuppressive activities. They can also mediate antitumor immune responses through changing their own phenotype and secreting related factors. Recent studies show that the infiltration of Th17 cells is associated with poor prognosis in breast cancer

. In contrast, infiltration of Th17 cells notably enhanced survival in lung cancer patients
[2]
. So Th17 cells are supposed as a double-edged sword in cancer. Further studies will be needed to advance our understanding of the role of Th17 cells in the immunopathogenesis of individual cancers. This article briefly reviews the differentiation and regulation of Th17 cells, the role and the latest progress of Th17 cells in the development of malignant tumors, in order to provide the improvement of more helpful ways for cancer immunotherapy.

effector T cells and play important roles in host defense against various pathogens and inflammatory pathogenesis. PI3K/AKT/mTOR signaling pathway can regulate cell metabolism, proliferation, differentiation, survival and other life processes, meanwhile, participate in the occurrence of tumors and autoimmune diseases. This review concentrated on the research progress in the regulation of the PI3K/AKT/mTOR signaling pathway on Th17 cells.
