Promotion effect of concentrated growth factor extracton on the proliferation and differentiationof gingiva-derived mesenchymal stem cells |

Abstract Objective To evaluate the effect of concentrated growth factors extract (CGFe) on the proliferation of gingiva-derived mesenchymal stem cells (GMSCs) and differentiation of F-actin cytoskeleton.Method GMSCs were cultured in vitro.Cells in the experimental group were cultured with α-MEM containing CGFe (10% FBS),while cells in the control group were cultured with α-MEM without CGFe (10% FBS).The proliferation of GMSCs cells was determined by MTT assay,the activity of alkaline phosphatase (ALP) was detected at 1,3,5 and 7 d after osteogenesis induction,the formation of calcium nodules was detected by alizarin red staining at 14 and 21 d,and the morphology of the F- actin cytoskeleton was observed by confocal laser scanning microscope (CLSM).Results A significant increase of absorbance at 1,3 and 5 d was showed in the experiment group compared to that in the control group,P<0.05.On the assessment of ALP activity of GMSCs,the absorbance of the experiment group at 1,3 ,5 and 7 d was significantly higher than that of the control group,P<0.05 or P<0.01.On the assessment of osteogenic differentiation activity,the mineralized nodules were detected by the alizarin red (Sigma) staining.The overall absorbance of the calcium nodules in the experimental group at 7,14 d was significantly higher than that in the control group,P<0.01.Conclusion CGFe can significantly promote the proliferation of GMSCsand osteogenic differentiation.
