Change of ALDH2 expression in mouse testicular mesenchymal cells induced by single heat stress |

Abstract Objective To investigate the role of ALDH2 in the reversibility of mouse testicular reproductive function impaired by single heat stress.Methods Eighteen C57 male mice were randomly divided into the heat stress group and the control group and immersed in water at 43℃ and 25℃,respectively,for 15 minutes.The heat treatment group was divided into 3 subgroups (n=3) on day 1,7 and 14 after treatment,and a control group (n=3) was set in each group.At 1,7,and 14 days of heat exposure,all the mice were sacrificed and their testis tissues collected for measuring the expression level of the ALDH2 protein by immunohistochemistry and Western blot.Results Immunohistochemistry showed that ALDH2 protein was expressed in Leydig's cells.Western blot showed that there was no significant difference in ALDH2 protein expression on the first day after thermal stress compared with the control group.Compared with the control group and the first day of thermal stress,ALDH2 protein expression was significantly increased on day 7 and 14 after thermal stress (P<0.01).There was no significant difference in ALDH2 protein expression between day 7 and Day 14 of thermal stress.Conclusion The expression of ALDH2 protein increased significantly in 7 days and 14 days after exposure.
