Effects of arsenic trioxide on the expression of p53, Bax and Bcl-2 in mice B16 cells |

Abstract Objective To investigate the growth inhibition role of arsenic trioxide(As2O3) on mice malignant melanoma B16 cell,and observe the expression of p53, Bax, Bcl-2 protein in B16 cell after induced by As2O3. To provide theory and experiment base for treatment of malignant melanoma. Methods The morphologic changes of B16 cells were detected by staining. The expression of p53, Bax, Bcl-2 protein was detected by Western blot. Results As2O3 caused morphologic change of B16 cells by staining. The expression of Bax and p53 protein was up-regulated in B16 cells by co-culture with As2O3(P<0.01). The expression of Bcl-2 protein was inhibited by As2O3(P<0.01). Conclusion As2O3 can cause morphologic change of B16 cells and promote apoptosis happening,which may be achieved by inducing the up-regulation of p53 and Bax protein expression, the down-regulation of Bcl-2 protein expression in B16 cells.
