A study on the expression and significance of HIF-1α on the maternal-fetal interface of fetal growth restriction |

Abstract Objective To study the expression of HIF-1α on maternal fetal interface between normal full-term newborns and fetal growth restriction (FGR) fetus and discuss the effect of HIF-1α and its target gene in the pathogenesis of FGR.Methods This study used classic ELISA method to detect the content of various substances in maternal blood,umbilical cord blood,cmaternal and child tissue surface of two groups by uniform linear calculation.Conducted a comparative analysis of the content of HIF-1α in poly formaldehyde fixed and wax embedded placental tissue of two groups,the test results were based on the content of HIF-1α calculated by standard curve,after the tissue sections being immunohistochemical stained and fluorescence stained,examine the expression of HIF-1α on the maternal surface and child surface.Microscope was used to observe the histological changes and the display intensity was determined by quantitative analysis by the image analyzer.The changes of inner organization structure in two sample cells were observed through an electron microscope, and the followed analysis is based on above mentioned observations.Results Immunohistochemical stains and immunofluorescence staining showed strong positive on the sub surface of FGR placenta.Compared the maternal surface of FGR with the maternal surface of normal full-term new born,the difference had no statistical significance;compared the placenta sub surface of FGR with the placenta sub surface of normal full-term, the difference had statistical significance;compared the maternal blood of FGR with the maternal blood of normal full-term fetus,the difference had nostatistical significance;compared the umbilical cord blood of FGR with the umbilical cord blood of normal full-term one, the difference had statistical significance.Conclusion The expression of HIF-1α on FGR placenta sub surface and the level of umbilical cord blood are significantly increasing by reducing the blood flow of the sub surface of placenta via inducing vascular endothelial injury, posing impact on the blood supply to the fetus,and induce chronic hypoxia, which plays an important role in the pathogenesis of FGR.
