Effects of HU-308 and nHA/PCL electrospun membrane on the biological properties of MC3T3-E1 |

Abstract Objective To explore the effects of different concentrations of HU-308 and nHA/PCL electrospun membranes on the biological properties of mouse pre-osteoblasts (MC3T3-E1).Methods The round electrospun membranes were placed at the bottom of the cell culture plates. MC3T3-E1 osteoblasts were inoculated and cultured in different concentrations of HU-308 (0, 10-10, 10-9, 10-8, 10-7, 10-6 mol / L). At 1, 4, and 7 days, cell proliferation and alkaline phosphatase (ALP) activity was detected; RT-qPCR was used to detect osteoblast-related genes OPG mRNA after cultured cells for 7 days. The morphology of the cytoskeleton after 24h was observed.Results After incubation for 1, 4, and 7 days, compared with that in the control group, the proliferation activity of cells in each experimental group was enhanced, P<0.05; with the extension of incubation time, the proliferative activity of cells in the same experimental group increased gradually, P<0.05. With the extension of incubation time, ALP activity in MC3T3-E1 cells in the same experimental group increased, P<0.05. At the same time point, compared with that in the control group, the low concentration of HU-308 (10-10 mol/L, 10-9 mol/L) promoted the expression of OPG mRNA, P<0.05. High concentration of HU-308 inhibited the expression of OPG mRNA, and the higher the concentration was, the more significant the inhibition was, P<0.05. The results of phalloidine staining showed that the cells in the 10-6 mol/L group had the largest spreading area and most explicit structure.Conclusion HU-308 and nHA/PCL electrospun membranes can promote the adhesion, proliferation, and differentiation of MC3T3-E1 cells.
