Abstract Objective To investigate the effect of miRNA-132 on proliferation and apoptosis of leukemia cell line K562, and explore its possible mechanism. Methods K562 cells were transfected with LipofectamineTM 2000.The expression of miRNA-132 was detected by real-time quantitative PCR.The effect of transfection on proliferation and apoptosis of K562 cells was detected by CCK-8 and flow cytometry. The protein expression levels of SRIT1 and P53 were detected by Western Blot. Results The expression of miRNA-132 in normal cells was significantly lower than that in K562 cells.Compared with the blank group and the negative control group, the expression of miRNA-132 was significantly increased in the transfected group.K562 cells’ proliferated and the apoptosis rate decreased significantly after transfection of miRNA-132 mimics.Compared with the blank control group and miRNA-132-NC group, after the high expression of miRNA-132,the expression of SIRT1 protein in the transfected group was significantly decreased, and the expression of P53 protein was significantly increased. Conclusion Highly expression of miRNA-132 significantly inhibits the proliferation of leukemia K562 cells and induces the apoptosis, which may be related to the enhancement of SIRT1 and P53 activity.
