Abstract Objective To study on the preparation process and anti-inflammatory activity of total flavonoids of PotentilladiscolorBge. Methods The plant was extracted with 70% ethanol, and purified with polyamide resin, and rutin was used as a control to investigate the distribution and purity of total flavones.Mononuclear macrophage RAW264.7 was induced with lipopolysaccharide (LPS), and after the treatment with different concentrations (25, 50, 100 μg·mL-1) of total flavonoids of PotentilladiscolorBge., quantitative PCR was used to detect the mRNA expression levels of inflammatory factors iNOS, IL-1β, and IL-6. Results The purity of total flavonoids of PotentilladiscolorBge. obtained by polyamide chromatography was 87.4%, and the yield was 82.6%. The total flavones of PotentilladiscolorBge. could significantly down-regulate the mRNA expression of inflammatory factors iNOS, IL-1β and IL-6 in RAW 264.7 cells induced by LPS. Conclusion The total flavonoids of PotentilladiscolorBge. can be prepared by polyamide resin chromatography, and its anti-inflammatory activity is related to the down-regulation of mRNA expression of iNOS, IL-1β, and IL-6.
